No lol. I've got quite a more effeminate (than masculine) sound to my voice anyway - I don't sound like a woman, but my tone is very soft. And so to answer your question, no I don't speak like a woman when I'm dressed in drag. The idea of a drag queen is that you're a man dressed as a woman, not trying to pull off being a woman
